Personality:
At first seen, Jo's just some girl in some bar that Sam and Dean roll into. However the Roadhouse isn't just any bar, and Jo isn't just some girl. The Roadhouse is a hangout spot for Hunters, and when Dean attempts to pick her up, she blows through him like he's not even worth her time. First time we ever see her, she's got a gun to his back. While he manages to disarm her, she does punch him in the face.
Jo is the daughter of Ellen and William Anthony Harvelle. William was a Hunter who worked with John Winchester, Sam and Dean's father, but was killed when she was younger. This is a very strong motivator. While her relationship with Dean certainly helps fuel the fire of wanting to be a Hunter, she's had this motivation as a child and grew up wanting to follow in his footsteps. This makes her a ridiculous amount of loyal.
At first she's seen as just an eager person, trying to put together cases and giving information to Sam and Dean. Her mother insists that she can't do this. However Jo is very much her mother's daughter. Stubborn and headstrong, eventually Jo pretty much tricks Dean into working with her on a job.
Part of her loyalty lies in the fact that she grew up not much differently than the Winchesters. One parent dead and with her mother running the Roadhouse, she had extensive knowledge on the supernatural. This loyalty is challenged when she found out exactly how her father was killed: John was not only the cause, but to save him more pain, he had to kill him himself.
While this news does change her, it really only made her more determined. When called on again, Jo still helps the Winchesters and even ends up dying trying to save them. She is a capable Hunter. She can carry a gun, knows how to play bait, and helps break into a house.
Her greatest weakness and asset is her willingness to get involved. Her inexperience does work against her, but being a hunter and hunter's daughter, she is resourceful. She will try. She nearly got caught with a the ghost in the wall, but even as she laid dying when the hell hounds got her, she had the idea of using a bomb to take out the dogs.
Due to her mother not allowing her to become a hunter, she does have a stubborn streak. She's sneaky but thorough about her information gathering, but this makes her charge into battle, somewhat naively. But she did eventually go off and start hunting on her own. She's resourceful like that.
In that sense she does know what she's talking about. Her research has always been impeccable. It's her handling of it that is lacking. She wants to save people. She became a hunter because of her father, but saving innocents is what is important to her, especially since most of the world does not know about demons or the supernatural. They have free reign on torturing and killing humans.
She does know how inexperienced she is, a lot of her actions are self serving in that manner. She wants to prove that she's just as tough and as good as the boys. After all, it's a male dominated work area. In this way, she also isn't that personable. It takes awhile to gain her trust. Getting hurt is an easy thing in their field. Their life isn't an easy one, but it's one she's chosen. She won't just sleep with Dean for this reason. Men came and went through the Roadhouse. She knows the game. It makes her come off a little sarcastic at times, protective of herself.
Jo believes what her mother told her about John using her father as bait. Regardless of what Meg who was possessing Sam at the time said, it doesn't matter. Meg is a demon, and they say horrible things and lie all the time. What she knows to be true is what Ellen told her. It shocked her at first, but once that was over, it wasn't dealt with. Sam and Dean weren't there. She has no reason to blame them.
Her relationship with her mother is the deepest. The two of them are all they have left. At first she's seen as more as a child, but she ends up telling her mother what she's going to do. She leaves and becomes her own Hunter. She's inherited much of her 'no shit taking' attitude, which is pretty funny to see Dean interacting with all of them. After she tricked him into working with her, he mentions something about Ellen and Jo asks sarcastically if he's afraid of her mother. He replies that he thinks so.
Her relationship with the Winchesters is also vital to who she is. It reminds her of her father, someone she mentions quite a few times in their company. But Dean and her definitely have some chemistry. Despite the fact she turned him down, there is something of a crush there - the constant smiles and protective nature. Possessed by the demon Meg, Sam taunts her about it.
She still stands strong. Even when presented with the fact that the night before Deluth, Minnesota may be her last night (and it was), she tells Dean that she'd rather spend the night with some self-respect than him. Which isn't necessarily an insult on him, just her ability to swat him down even when he's hitting on her as hard as he is. Just like her mother, she doesn't take it from anyone.
She's also loyal to Ash, a Lynard Skynard look alike who turns out to be a genius. He lives at the Roadhouse, supplying information to the Hunters. He is a genius and she defends him to both Sam and Dean. However he dies early on when the Roadhouse is destroyed so we never get to see exactly how close they are. In a future episode, in heaven Ash finds out that Jo and Ellen had died and seems pretty beat up about the news.
Abilities: Jo's a pretty average human when it comes to abilities, however she is a Hunter, which means she knows how to fight and knows how to use various weapons. She's also knowledgeable in the occult and the supernatural.

[She looks around, still a little shaken from the whole being teleported thing. There's a look of disbelief.] Gotta say, if this is the afterlife? Guess I was expecting more gardens and sunshine. You know, the after school special type of 'Heaven'. You think you know a thing or two when you know an angel.
[Jo looks around.] So where am I exactly? And has anyone seen my mom? Ellen Harvelle. Or the Winchesters? Whatever this is, there's bound to be one of them here. [She purses her lips, giving a look that said 'they're always involved'.]
Ugh, and a new pair of clothes? [The ones she's wearing have her blood smeared all over them. It's a little jarring to look down and not expect her guts to fall out. That's going to take a moment or two to set in. She's not dead. That may not be a good sign either. She looks back up.]

So some big bads were out in the city, this weird city that she was still trying to process. Running wasn't a thing Jo often did. But she had a gun. That was a start at least. Some knew place opened up. Why wasn't she surprised?
Well now she was heading toward the other side of the city. Dean and Sam were heading there, a few others she'd met a long the way. They were going to take these things down, the Hunter way. Which meant going balls to the wall and taking it however they could. They didn't exactly have the advantage, but that was rarely the case back home. What would make that different here?
Despite the fact she was dead, she'd never stopped. Call it the Harvelle attitude. But being here just meant there were other bad things to take down. It was good to be with the Winchester. All things considered with their own fates, it was good to be with family.
Sam and Dean were always Sam and Dean. It was nice having something to rely on. Cas was there. Sam and Dean's parents were. It was a little-- jarring to see John Winchester, and she knew part of her would want to talk about her own father, but right now there were bigger things. Things that could result in them not talking. Which was a priority a lot here it seemed.
But there wasn't much to do. Except keep trying. She'd never stop with that. If that meant fighting or just being there. So as she turned the corner, the hunter's daughter cocked the gun. Jo was raring to go.
